Factors Affecting Production of ¥â - 1,3 - Glucanase from Bacillus sp .

Abstract

|
A bacterium capable of producing ¥â-1,3-glucanase was isolated from soil, and identified as a strain of Bacillus. Effects of culture conditions and medium compositions on enzyme production were investigated. Among tested carbon sources, cudlan(¥â-1,3-glucan) was most effective for production of the enzyme, whereas mono- and di-saccharides were generally ineffective. Therefore it seemed that the enzyme was induced when ¥â-1,3-glucan was used as a sole carbon source. The optimal concentration cudlan for the enzyme production was 7.5%. The enzyme was also induced by baker¢¥s yeast cells as a nitrogen source, and repressed by glucose in the culture medium. The enzyme was maximally produced by cultivating the isolated Bacillus sp. at initial pH 6.0, and 32¡É.
